目的通过网络药理学的方法预测气滞胃痛颗粒抗炎镇痛主要活性成分的作用靶点,结合中医方解配伍理论对其多成分-多靶点-多通路的作用进行分析。方法基于TCMSP中药系统生物学分析数据库收集气滞胃痛颗粒中6味中药的主要化学成分,并借助LC-MS技术对所筛成分进行分析,通过TCMSP检索和Pharmmapper软件预测获取各成分主要的作用靶标,并通过DIP数据库,利用蛋白质相互作用信息建立药物靶标与炎症疼痛靶标的关联,构建药物-靶标-疾病网络,通过网络特征分析气滞胃痛颗粒抗炎镇痛的作用靶标,阐释其抗炎镇痛的主要作用机制。结果根据网络分析,共有44个炎症疼痛靶点与气滞胃痛颗粒密切相关,其中直接作用靶点有20个,主要是对环加氧酶-2(COX-2)和诱导型一氧化氮合酶(i NOS)等蛋白酶的作用,作用机制可能与调节肿瘤坏死因子(TNF)信号通路、NOD样受体(NLR)信号通路、血管内皮生长因子(VEGF)信号通路等与炎症疼痛密切相关的信号通路有关。结论气滞胃痛颗粒抗炎镇痛作用体现了中药多成分、多靶点、多途径的作用特点,该研究为深入阐释气滞胃痛颗粒抗炎镇痛作用机制提供科学依据,并且进一步说明了中医药古方配伍理论的科学性。  相似文献

目的 制备蕨麻素冻干脂质体注射剂并研究其理化性质。方法 采用薄膜超声分散法并冷冻干燥来制备蕨麻素脂质体,用葡聚糖凝胶(Sephadex G-50)微型柱离心法测定包封率。结果 制得的蕨麻素脂质体包封率为56.62%,粒径范围为71.70—166.47nm,透射显微镜下观察脂质体为粒径均匀的球类或近球状小囊泡。结论 蕨麻素脂质体处方和制备工艺稳定可行,重现性好。  相似文献

The parasitic copepod Sinergasilus major is an important pathogen of grass carp Ctenopharyngodon idella. To understand the immune response of grass carp to the copepod infection, suppression subtractive hybridization method was employed to characterize genes up-regulation during the copepod infection in liver and gills of the fish. One hundred and twenty-two dot blot positive clones from infected subtracted library were sequenced. Searching available databases by using these nucleotide sequences revealed that 23 genes are immune-related, including known acute-phase reactants, and four novel genes encoding proteins such as source of immunodominant MHC-associated peptides (SIMP), TNF receptor-associated factor 2 binding protein (T2BP), poliovirus receptor-related protein 1 precursor, glycoprotein A repetitions predominant (GARP). The differential expression of seven immune genes, i.e. GARP, alpha-2-macroglobulin, MHC class I, C3, SIMP, T2BP, transferrin, as a result of infection was further confirmed by RT-PCR, with the up-regulation of alpha-2-macroglobulin, MHC class I, C3, SIMP and T2BP in the liver of infected fish, and down-regulation of SIMP in the gills of infected fish. The present study provides foundation for understanding grass carp immune response and candidate genes for further analysis.  相似文献

目的:探究细胞因子TNF-α、IL-6和IL-8在白血病异基因造血干细胞移植后的变化和病情及预后的关系.方法:用放射免疫分析检测了白血病异基因造血干细胞移植患者(48例)、正常健康人(30例)血清TNF-α、IL-6和IL-8水平,对移植后发生或未发生急性移植物抗宿主病(aGVHD)及发生或未发生慢性移植物抗宿主病(cGVHD)进行了比较,并对预后也进行比较.结果:急、慢性GVHD组与未发生急、慢性GVHD组患者的血清TNF-α、IL-6及IL-8水平均高于正常对照组(P<0.05,除未发生cGVHD组的TNF-α浓度低外);急、慢性GVHD组与未发生急、慢性GVHD组比血清TNF-α、IL-6及IL-8水平均升高(P<0.05);移植后死亡组生存时间≤6月及>6月(除>6月的IL-8水平外)各组TNF、IL-6与IL-8水平均高于同期的存活组(P<0.05).结论:异基因造血干细胞移植术后动态监测TNF、IL-6、IL-8水平变化有助于对发生aGVHD 或cGVHD的可能提供另一种检测方法,并对判断愈后有一定的临床意义.  相似文献

目的 研究甘草中有效组分对脓毒血症的防治作用及机制。方法 采用小鼠骨髓来源巨噬细胞(bone marrow-derived macrophages,BMDM)构建体外NOD样受体热蛋白结构域相关蛋白3(NOD-like receptor thermal protein domain associated protein 3,NLRP3)炎症小体激活模型及H2O2诱导的氧化应激模型筛选出甘草抗炎、抗氧化应激的有效组分,采用ip脂多糖(lipopolysaccharide,LPS,20 mg/kg)诱导C57BL/6小鼠脓毒血症致死模型以及脓毒血症模型,评价致死模型中小鼠的生存率及脓毒血症模型小鼠腹腔灌洗液中巨噬细胞和中性粒细胞在渗出细胞中的占比以及外周血和腹腔灌洗液中白细胞介素-1β(interleukin-1β,IL-1β)及肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平。结果 基于BMDM细胞NLRP3炎症小体激活模型筛选出甘草中有效组分刺甘草查耳酮,可显著抑制半胱氨酸天冬氨酸蛋白酶-1(cystein-asparate protease-1,Ca...  相似文献

目的探讨精准护理在新型冠状病毒肺炎流行期间肿瘤晚期患者中的应用效果。方法选取2020年1—3月于医院肿瘤科住院的82例肿瘤晚期患者作为研究对象,所有患者住院后均实施精准护理,比较患者在入院和出院时的焦虑自评量表(SAS)及抑郁自评量表(SDS)评分。结果出院时,患者的SAS及SDS评分均低于入院时,差异有统计学意义(P<0.05)。结论精准护理可有效降低新型冠状病毒肺炎流行期间肿瘤晚期患者的焦虑及抑郁水平。  相似文献

2010年7月下旬历史罕见的水灾席卷巴基斯坦,全国四分之一土地被淹,2 000余万人受灾。中国国际救援队两批次78人赴巴基斯坦开展洪灾救援。这是中国国际救援队成立近10年以来最大规模的一次救援,历时最长、派出人员最多、完成救治病人数量最多,先后救治2万余名灾区病人,圆满完成了医疗救援和卫生防疫任务。在总结以往国际救援护理经验基础上,结合洪灾现场实际,将全科护理管理模式应用于此次洪灾救援实践中,收到了良好效果。  相似文献

Polybrominated diphenyl esters are emerging environmental contaminants with few toxicological data, being a concern for the scientific community. This study evaluated the effects of BDE-47 on the health of Oreochromis niloticus fish. The animals were exposed to three doses of BDE-47 (0, 0.253, 2.53, 25.3 ng g−1) every 10 days, for 80 days. The BDE-47 affected the hepatosomatic and gonadosomatic index in female and the condition factor by intermediate dose in both sexes. The levels of estradiol decreased and the T4 are increased, but the vitellogenin production was not modulated in male individuals. Changes in AChE, GST, LPO and histopathology were observed while the integrated biomarker response index suggests that the lowest dose of BDE-47 compromised the activity of antioxidant enzymes. The oral exposure to BDE-47 in environmental concentrations is toxic to O. niloticus and the use of multiple biomarkers is an attribution in ecotoxicology studies and biomonitoring programs.  相似文献

ObjectivesIn a clinical setting, patients have been observed to complain of discomfort and to discontinue treatment because of chemotherapy-induced peripheral neuropathy. This experimental study was conducted to determine the effect of a salt-water bath in the management of chemotherapy-induced peripheral neuropathy.MethodOne hundred and three patients who received taxane and platinum-based chemotherapy due to cancer and developed peripheral neuropathy associated with the treatment between December 2018 and June 2020 were included in the study. The patients were assigned to the control and experimental groups (1-warm salt-water and 2-cold salt-water) following the randomization checklist. While control groups did not receive any interventions, the patients in the salt-water group were asked to apply warm (41 °C) or cold-water (23–26 °C) baths to their hands/feet for 30 min every other day for 14 days. The data were collected at the beginning of the study and at the end of its first and second weeks using the Patient Information Form and National Cancer Institute (NCI)-CTCAE v5.0 toxicity criteria as well as the EORTC QLQ-C30 and EORTC QLQ-CIPN20 quality of life scales.ResultsThe patients had a mean age of 55.6 ± 10.3, and most of them were treated following a breast cancer diagnosis. At the beginning of the study, Grade 3 peripheral neuropathy severity and quality of life scores of the cold/warm salt-water and control groups were similar. Due to repeated follow-ups, it was determined that the peripheral neuropathy severity decreased and the quality of life scores increased statistically significantly in the patients in the cold salt-water bath group compared to the control group.ConclusionThis study's results suggest that a cold salt-water bath can be an effective approach in managing the development of peripheral neuropathy due to taxane and platinum-based treatment.  相似文献
